What it is

The Olympus DSU is a microscope used in metrology and inspection work. The DSU can be configured on an IX81 stand with a motorized Z drive, a filter cube, an objective turret, a Prior arc lamp and xenon light source, and a Windows 7 computer running Slidebook.[1][2]

How it works

The DSU uses a spinning disk with a pattern of slits to create a virtual pinhole as the disk rotates. The DSU is designed to improve the balance between confocality and light throughput.[3]

The DSU uses fluorescence excitation light from a white light source such as a xenon or mercury lamp. The excitation light is filtered, reflected through the spinning slit disk, passed through the objective, and directed onto the specimen, while emitted fluorescence returns through the disk for wavelength selection and image formation.[4]

Applications

The DSU is used for basic confocal Z-stack imaging. The DSU is also used with fluorescence excitation for live-cell confocal microscopy.[3][4]
